With the deadlock unable to be broken, Semenyo replaced Athletic Bilbao forward Iñaki Williams, a game-changing move.
He went close as he raced into the box, moving the ball to his left-foot and drilling just over the bar from a tight angle but he went one better in the sixth minute of stoppage time.
In what was surely the last attack of the game, a Ghana free-kick resulted in a penalty box scramble and Semenyo was on hand to power home the loose ball, his first competitive goal for his country.
Ghana will travel to Talatona to take on Angola again on Monday as they look to consolidate their control on Group E, of which they're currently top.
Meanwhile, Philip Billing was a second-half substitute as Denmark cruised to victory over Finland in their opening UEFA EURO 2024 qualifier.
The scores were level at 1-1 when Billing came on in the 77th minute, Rasmus Højlund's opening goal had been cancelled out by Oliver Antman.
Billing's addition, which is just his fourth international cap, had an impact as Denmark took hold of the game and Højlund completed his hat-trick with two further strikes.
The Cherries midfielder could next be in action on Sunday when they take on Kazakhstan.
